Specificity | MGR48 binds to proteins localised within the secretory granules of the subventral gland in G. rostochiensis, and has also been found to react with the subventral glands of H. glycines. MGR48 has been shown to recognise six subventral gland proteins (svp3Q, svp31a, svp3i, svp32, svp39 and svp49) by Western blotting. |

Immunogen | Sixteen mice were immunised intraperitoneally with isolated protein fractions from homogenised preparasitic second-stage juveniles of G. rostochiensis, which were mixed 1:1 with Freund's incomplete adjuvant. weeks later antiserum samples were collected for immunofluorescence microscopy. One mouse that was selected for monoclonal antibody production received a final booster injection 12 weeks after the first immunization. Three days later the mouse was sacrificed, and MAb-producing hybridoma cell lines were obtained by fusing spleen cells with SP 2/0 myeloma cells. |
Application Notes | MGR48 has been used in Western Blot analyses in order to confirm the presence of secretory components in collected G. rostochiesis material (Robertson 1999). Further, MGR has been employed to immunopurify the endoglucanases from G. rostochiensis and H. glycines (Smant 1998). MGR48 has also been used in immunofluorescence testing to compare DMT-treated and nontreated preparasitic PRD-hatched J2 (Smant 1997). |
