Specificity
This antibody recognizes and binds the Ku protein heterodimer (Ku70/80), a nuclear protein that is also present on the surface of some tumor cells.

Immunogen
The original antibody was isolated from a naïve phage antibody library n-CoDeR®. First substraction was done by incubation with the T cell leukemia cell line MOLT-4. Selections were performed on the pancreatic adenocarcinoma cell line PL45, during 1 h incubation at 37° C, allowing internalization of the phage scFv-receptor complexes. Internalized phages were retrieved by lysing surface-stripped cells with 100 mM triethylamine.
Application Notes
Ku protein is a heterodimer of 2 tightly associated sub-units called the Ku70 and Ku80, normally located in the nucleus, where it is involved in nonhomologous end-joining processes, which is responsible for repairing double stranded DNA breaks. The receptor-mediated endocytosis of Ku70/80 is rapid (t1/2 12 min) and extensive (90% of the receptor pool inside the cell after 100 min) as measured by rotating radioimmunoassay. Ku70/80 was also successfully used as a port of entry for cytotoxic payloads to tumor cells of various origin. INCA-X recognizes and binds Ku protein heterodimer. The binding specificity of INCA-X for Ku70/80 was confirmed with immunoprecipitation and flow cytometry. This antibody is capable of internalization and has the potential for use in immunotherapy. The internalization of INCA-X in pancreatic carcinoma cells was confirmed using confocal microscopy. The uptake, internalization and retention of this antibody was studied using a rotating radioimmunoassay. Carcinoma cells were treated with INCA-X and subsequently incubated with a saporin toxin-conjugated anti-human IgG antibody (Hum-ZAP) to determine the immunotoxin mediated cell cytotoxicity.
