Description

The antibody has a proven and strong Biotin-Streptadvidin/HRP staining at a 1/2000-1/4000 dilution in rat amygdala, and spinal cord.
The specificity of the antiserum was evaluated by soluble pre-adsorption with the peptides in question at a final concentration of 10-5M. CGRP immunolabeling was completely abolished by pre-adsorption with rat α -CGRP and partially eliminated by pre-adsorption with rat Ÿ -CGRP. Pre-adsorption with the following peptides resulted in no loss of immunostaining: rat amylin, rat adrenomedulin, calcitonin, neurotensin, somatostatin, substance P, leucine enkephalin, methionine enkephalin, VIP, CCK-8, vasopressin and neuropeptide Y.

APPLICATION

Quality Control

The antibody has significant fluorescein staining at a 1/100–1/200 dilution and significant Biotin/avidin-HRP staining at a 1/2,000–1/4,000 dilution in rat amygdala, and spinal cord. The specificity of the antiserum was evaluated by soluble preadsorption with the peptides in question at a final concentration of 10-5 M. CGRP immunolabeling was completely abolished by preadsorption with rat a-CGRP and partially eliminated by preadsorption with rat b-CGRP. Preadsorption with the following peptides resulted in no loss of immunostaining: rat amylin, rat adrenomedulin, calcitonin, neurotensin, somatostatin, substance P, leucine enkephalin, methionine enkephalin, VIP, CCK-8, vasopressin and neuropeptide Y.

Perfusion Fixation

• Fixative: 4% paraformaldehyde in 0.1 M phosphate buffer, pH 7.4; 500 mL over 20-30 min.
• Post Fixation: 1.5 hr. at 4°C in 4% paraformaldehyde in 0.1 M phosphate buffer, pH 7.4.

Special Instructions

It is recommended that the researcher perform a primary antibody dilution series using our dilution recommendations as a guideline. Note that a change in the fixation or buffering system from our protocol may change the configuration of the protein which could alter the reactivity with the tissue tested.

Not applicable. Antibody concentration is only relevant for purified antibodies.

Storage

After reconstitution, use immediately or refrigerate at 2º–8ºC up to 2 days. For long term storage, aliquot and freeze at -15°C or lower. Avoid repeated freeze/thaw cycles.
