Specificity | The antibody is highly selective for Tetrabromobisphenol A and it shows negligible cross reactivity (<0.1%) with other tested analogues (TBBPA-bAE, TBBPA-bOHEE, DBDPE, HBCD, BDE-47, BDE-99, BDE-100, BDE-153, BDE-154, BDE-183, 5-OH-BDE 47, 6-OH-BDE 47, BPA). Tetrabromobisphenol A is a ubiquitous brominated flame retardant. |

Immunogen | The original antibody was generated by immunizing an alpace with a TBBPA hapten coupled to thyroglobulin. A VHH phage display library was constructed with the blood lymphocytes and panning against heterogeneous coating antigens. |
Application Notes | The VHH fragment was used to develop an ELISA for monitoring environmental and human exposure to TBBPA. This assay had an IC50 for TBBPA of 0.40 ng/mL. Thermal stability studies showed the VHH−antigen binding signal retained about 80% and 20% of the unheated control signal after heating at 90 °C for 10 and 90 min, respectively. The recoveries of TBBPA from spiked soil and fetal bovine serum samples showed good correlation with liquid chromatography−tandem mass spectrometry method (Wang et al, 2014; PMID: 25068372). The VHH fragment was fused with alkaline phosphatase (AP) and a one-step immunoassay for the detection of TBBPA was developed, showing a half-maximum signal inhibition concentration (IC50) of 0.20 ng/mL. Compared to the parental VHH, the VHH-AP was able to bind to a wider variety of coating antigens and the assay sensitivity was slightly improved (Wang et al., 2015; PMID: 25849972). |
