Specificity | This antibody binds a conformational epitope on the A subunit of Shiga-like toxin 2 comprising region A (StxA2 residues 42 to 49), region B (StxA2 residues 96 to 100), and region C (StxA2 residues 244 to 259) (PMID: 19433543). This antibody does not bind A subunit of Stx1. StxII is a toxin produced by Shiga toxin (Stx)-producing E. coli (STEC) strains. Infections with STEC can lead to diarrhea, hemorrhagic colitis, and hemolytic uremic syndrome (HUS). |

Immunogen | The original antibody was generated by immunizing 4 to 8 week old female BALB/c mice with 25µg of formalinized StxII toxoid or 25µg of glutaraldehyde treated toxoid in complete Freund adjuvant. |
Application Notes | The binding characterization of the original IgG1 version of this antibody towards Shiga like toxin 2 was done using ELISA. The original IgG1 version of this antibody was also used to immunoprecipitate SLT-Il toxoid. This antibody is capable of neutralizing the cytotoxicity of SLT-ll but not of SLT-I, Shiga toxin, or a variant of SLT-II produced by E. coli that causes edema disease of swine (PMID: 3053764). This antibody was also used a primary antibody in the detection of SLT-II in a western blot assay. It was reported that binding of 11E10 to Stx2 prevented the toxin from inhibiting protein synthesis in an in vitro assay but also altered the overall cellular distribution of Stx2 in Vero cells. It was suggested that binding of antibody 11E10 to Stx2 neutralizes the effects of the toxin by preventing the toxin from reaching and/or inactivating the ribosomes (PMID: 19433543). |
