Specificity | This antibody is specific for the B subunit of Shiga-like toxin 1. This antibody does not bind B polypeptide of Stx2. Stx 1 is a toxin produced by Shiga toxin (Stx)-producing E. coli (STEC) strains. Infections with STEC can lead to diarrhea, hemorrhagic colitis, and hemolytic uremic syndrome (HUS). |

Immunogen | The original antibody was generated by immunization of BALB/c mice (female, 6 to 8 weeks old) with purified, biologically active Shiga-like toxin (SLT) from E. coli H30. |
Application Notes | The original IgG1 antibody neutralized the cytotoxicity of E. coli Shiga-like toxin (SLT) and were able to immunoprecipitate intact labeled toxin with Staphylococcus aureus protein A. The original IgG1 antibody neutralizes Shiga toxin from Shigella dysenteriae type 1 and Shiga-like toxic activities in crude cell extracts from Shigella flexneri, Vibrio cholerae, Vibrio parahaemolyticus, and Salmonella typhimurium. The IgG1 version of this antibody was also used in a colony blot assay for the detection of bacterial colonies which produce high levels of SLT (PMID: 3905611). This antibody neutralizes the cytotoxic and lethal activities of Stx1, This antibody was also reported to block the binding of Stx1 to its receptor, globotriaosyl ceramide by binding three nonlinear regions of the molecule that span receptor recognition sites on StxB1, one of which includes the essential residue 55N. This antibody was able to identify histidine- tagged StxB1 in a western blot assay. It did not recognize Stx1d (PMID: 17030576). |
