Immunogen | The original antibody was generated by immunizing rabbits with recombinant phosphorylated peptide pPLC-γ(RNPGF-pY-VEANPGGGSC) conjugated with KLH as an immunogen. |
Application Notes | The binding reactivity of this antibody to recombinant phosphorylated pPLC-γ peptide was confirmed using ELISA. The binding reactivity was further confirmed using western blot, of phosphorylated PLC-γ from NIH3T3 cell lysates. The scFv version of this antibody was used fused to human IgG1 using a suitable linker for the generation of scFv-Fc mini-antibody. This minibody was capable of immunoprecipitating pPLC-γ. The fluorescent scFv version of this antibody, when expressed intracellularly was reported to inhibit signal transduction by PLC-γ in transfected into NIH3T3 cells as observed using fluorescent microscopy (US20090233358). |
