This antibody was raised by immunizing CAF mice with human thymocytes.
Application Notes
CD8+ T cell effector function assays were preformed using this antibody. The cell lines used for this study are HLA A*0201-restricted CD8+ T cell clones: 1) ILA1, specific for the human telomerase reverse transcriptase-derived epitope ILAKFLHWL (residues 540–548) (30, 31); 2) ALF3, specific for the influenza A matrix protein (M1)-derived epitope GILGFVFTL (residues 58–66); and 3) MEL5 and MEL187.c5, specific for the Melan-A–derived epitope ELAGIGILTV (residues 26–35) (32). The HLA B*0801-restricted CD8+ T cell clone LC13 is specific for the EBV EBNA3A-derived epitope FLRGRAYGL (residues 339–347) (33), and the HLA B*3508-restricted CD8+ T cell clone SB10 is specific for the EBV BZLF1-derived epitope LPEPLPQGQLTAY (residues 52–64) (34). The HLA DR*0101-restricted CD4+ T cell clone C6 recognizes the influenza A hemagglutinin-derived epitope PKYVKQNTLKLAT (residues 307–319). CD8+ T cell lines specific for the EBV BMLF1-derived epitope GLCTLVAML (residues 280–288). (Clement et al, 2011; pmid:21677135). Lysis of Lymphoid Populations was tested with this antibody and rabbit complement (Reinherz et al, 1980; pmid:6966400). Human thymocytes and peripheral T cells were analyzed using flow cytometry with this antibody (Reinherz et al, 1980; pmid:6966400). Human CTLs were used for flow cytometry with this antibody (Wooldridge et al, 2003; pmid:14662868). Human CD8+ T lymphocytes and NK cells were used in an apoptosis study with this antibody (Contini et al, 2003; pmid:12594841). A cytotoxic assay was preformed on CTLs with this antibody. The assay used was a a standard 4-h 51Cr-release assay (Contini et al, 2003; pmid:12594841). This antibody was used for an immunofluorescence experiment on fresh human peripheral mononuclear blood cells (PBMC) (Gattass et al, 1988; pmid:3258844). A competition assay was preformed on PBMC with this antibody. part of this assay was a flow cytometry experiment (Gattass et al, 1988; pmid:3258844). A standard 4h 51 chromium release assay was used to evaluate cytotoxic activities of CTLs. In the assay this antibody was used (Hermann et al, 1993; pmid:8103147).
