Specificity
This antibody is specific for the α5 integrin subunit, and recognises a cation-independent, ligand-induced binding site epitope. Integrins are heterodimeric integral membrane glycoproteins, which are expressed by a range of cells, including T cells, NK cells, fibroblasts and platelets, where they mediate cell adhesion. Integrin α5β1 acts as a receptor for fibronectin and fibrinogen, and is involved in cellular adhesion to fibronectin. This antibody recognises a specific conformation of the α5β1 integrin associated with integrin translocation and fibronectin matrix formation.

Application Notes
The specificity of this antibody for α5 integrin has been confirmed by ELISA using immobilised α5β1 integrin (Clark et al, 2005). This antibody is conformation-dependent, and is able to convert α5β1 integrin into a ligand-competent form and promote fibronectin binding. In a K562-cell adhesion assay, this antibody has been shown to stimulate α5β1 integrin to bind fibronectin, and, by extension, promote cell adhesion (Clark et al, 2005). The ability of this antibody to stimulate α5β1 integrin function has been confirmed in solid-phase ligand-binding assays (Clark et al, 2005). This antibody has been used to specifically stain integrin localised to fibrillar adhesions in indirect immunofluorescence assays (Clark et al, 2005). It has also been used as an activation reporter to allow the early endocytic steps of active α5β1 integrin in endocytic cells to be assessed in fluorescent confocal analysis and immunofluorescence analysis (Valdembri et al, 2009).
