Specificity
The antibody sterically blocks the arginase 1 active site near the exterior surface of the enzyme. Approximately 80% of all complexes identified were composed of two arginase 1 trimers and three antibodies, forming a 2 3 complex, while the remaining 20% of complexes were composed of two arginase 1 trimers and two antibodies; however, the epitope and paratope interactions between each antibody and arginase 1 monomers are identical. Arginase is a manganese-containing enzyme that catalyzes the hydrolysis of L-arginine to L-ornithine and urea, and modulates T-cell-mediated immune response.

The antibody is a human antibody identified via yeast display technologies and isolated after panning against arginase 1.
Application Notes
The structure and epitope mapping of the complex arginase antibody was determined by cryo-electron microscopy. The antibody was assessed for its potency and extent of inhibition of human arginase using an assay that detects the formation of thioornithine from thioarginine. In this assay, as well as an LCMS assay, it was determined that the antibody could fully inhibit the enzymatic activity of arginase. To assess the kinetic mechanism of inhibition of the antibody, increasing fixed concentrations of antibody were tested across a range of thioarginine concentrations. The Ki for the antibody against human Arg1 was 3.3 ± 0.3 nM (Palte et al., 2021; PMID: 34326456).
