The original antibody was prepared by immunization of Llama glama with whole gliadin.
Application Notes
This antibody specifically binds an epitope unique for wheat alpha gliadins which is not present in gamma or omega gliadins. It does not cross reat with rye or barley gliadins. VHH26 can detect gliadins in denaturing conditions and it retains its binding capacity in presence of 15% ethanol, 0.5% 2-mercaptoethanol, and 0.5 M guanidinium chloride. Capture ELISA using VHH26 was able to detect gliadins in samples shown as negatives by conventional ELISA. This antibody can be used for detection of gluten in commercial food samples using ELISA (limit of detection 0.24 ppm) and by competitive ELISA using conventional antibodies (limit of detection: 1 ppm) (PMID: 20039674).
