Specificity | This antibody binds human monomeric and high molecular weight forms of wild-type a-syn.Alpha-synuclein is a small 140 amino acid presynaptic neuronal protein, a major component of Lewy bodies and a member of the synuclein family. Misfolding, abnormal accumulation, and secretion of α-Synuclein (α-Syn) are closely associated with synucleinopathies, including Parkinson’s disease (PD). α-syn is abundant in the brain, and smaller amounts are found in the heart, muscles, and other tissues. In the brain, α-syn is found mainly at the tips of neurons in the specialized structures known as presynaptic terminals and has been shown to comprise up to ∼1% of total proteins in the neuronal cytosol. Recent evidence suggests that α-synuclein (α-syn) can contribute to the pathogenesis of amyotrophic lateral sclerosis (ALS). |

Application Notes | This antibody is capable of binding recombinant human alpha-synuclein in an ELISA.The intracellular binding of scFv D10 antibody to α-syn was studied by cotransfection of COS-7 cells with a D10-HA-NLS plasmid (hemagglutinin (HA) tag to detect scFv; nuclear localization signal (NLS) for retargeting to nucleus) and an α-syn plasmid. Immunocytochemical staining of HEK 293/α-syn/D10 cells reveals that the D10 intrabody has a predominantly cytoplasmic distribution and does not induce visible α-syn aggregate/inclusion formation. Transfection of D10 scFv gene into an HEK 293 cell line that overexpresses wild-type α-syn, showed that the D10 intrabody stabilizes detergent-soluble monomeric α-syn and inhibits the formation of detergent-insoluble high-molecular-weight α-syn species. It was seen that this antibody also ameliorated decreased cell adhesion characterized by α-syn-overexpressing cells (PMID:15564134). This antibody is capable of detecting total α-syn in a western blot (PMID: 26287258). |
