Specificity | This antibody binds SN-38, which is an active metabolite of irinotecan (a semisynthetic analog of camptothecin - a topoisomerase I inhibitor) generated by carboxyl esterases through hydrolysis. SN-38 has potential antineoplastic activity. It binds and inhibits topoisomerase I by stabilizing the cleavable complex between topoisomerase I and DNA, resulting in DNA breaks, inhibition of DNA replication, and apoptosis. In vitro cytotoxicity assays have shown that SN-38 is 1,000-times more cytotoxic than irinotecan against various cancerous cells. SN-38 is commonly used as a cytotoxic drug payload in the ADC field. |
