Specificity  | This antibody binds human interleukin-13 at the surface of helices A and D close to N- and C- termini of the IL-13 polypeptide chain. IL-13 is a cytokine secreted by T helper type 2 (Th2) cells, CD4 cells, natural killer T cell, mast cells, basophils, eosinophils and nuocytes. It inhibits the production of inflammatory cytokines (IL-1, IL-6, TNF, IL-8) by LPS-stimulated monocytes. It synergizes with IL2 in regulating interferon-gamma synthesis. It is a mediator of allergic inflammation and different diseases including asthma.  |

Immunogen  | The original mouse antibody m836 was generated by immunizing BALB/c mice with a natural variant of human IL-13 known as R130Q. The humanized version H2L6 was generated by grafting CDRs of mouse antibody onto selected human heavy chain and light chain frameworks and finally site-directed mutagenesis of two methionine in the CDRs of heavy chain with binding affinity optimization resulted in the generated of M1295.  | 
Application Notes  | This antibody can bind human IL13 in an ELISA. The affinity of original IgG1 antibody for wild type human IL-13 was determined by BIAcore to be 5.7 ± 1.3 pM (PMID: 20226193). Preclinical studies confirm the functional effects of M1295-mediated neutralization of human IL-13 in human cell based in vitro assays using mechanistically relevant endpoints associated with the pathogenesis of asthma, atopic dermatitis, and eosinophil-mediated disease (US8399630).  |
