Application Notes | The original mouse antibody was able to bind fibrin even in the presence of human fibrinogen at the concentration found in plasma. This antibody when intravenously administered in a chicken ex vivo circulatory model bound human fibrin-coated disks placed in an extracorporeal chamber (PMID: 6648524). An immunoconjugate (tPA-59D8) generated from tissue plasminogen activator (tPA) and anti-fibrin antibody 59D8 degraded less fibrinogen and consumed less alpha 2-antiplasmin than did tPA alone, suggesting that more potent and more selective thrombolysis can be achieved by efficiently directing tPA to the site of a thrombus (PMID: 3118374). This antibody was used in the generation of a single-chain urokinase plasminogen activator (scu-PA) and -59D8 Fab' conjugate which had a fibrinolytic potency 33-fold more than that of tissue plasminogen activator (PMID: 2111744). A recombinant r-scuPA(32kDa)-59D8 was 6 times more potent than high molecular mass scuPA in lysing a human plasma clot in vitro and was 20 times more potent than high molecular mass scuPA in the rabbit jugular vein model of thrombolysis. |
