Application Notes | FGF-1 plays an important role in the regulation of cell survival, cell division, angiogenesis, cell differentiation and cell migration. Studies have shown that FGF-1 is overexpressed in the early stages of several kinds of cancer. Functional analysis showed that the overexpressed scFv1C9 in MCF- 7 cells targeted endogenous FGF-1 and prevented the translocation of FGF-1 into the nucleus, resulting in the blockade of the intracrine pathway of FGF-1, which caused the G1 arrest by p21 up-regulation. Binding of scFv1C9 to FGF-1 was confirmed with ELISA. The antigen-binding ability of scFv1C9 with endogenous FGF- 1 was confirmed in vivo by using of an immunoprecipitation assay. FGF-1 immunohistochemical staining of breast ductal carcinoma tissues was used to demonstrate that FGF-1 was increasingly expressed in breast cancer tissues.(PMID:21360642). |
