Immunogen
The original antibody was isolated from a non-immune semisynthetic llama library constructed from blood samples (6 ml) collected from the left jugular vein of three research naïve mature llamas.
Application Notes
The binding specificity of this antibody towards cholera toxin was checked using ELISA (PMID:17165813). SPR was used to determine the dissociation constant (Kd) of VHH A9; the Kd was determined to be 77 ± 5 nM (PMID: 23385744). It was reported that two point mutations (N27D and R71I) help stabilize A9 wild-type form which has a relatively low melting point (PMID: 31342597).

A semisynthetic llama library was constructed from non-immune llamas and binders specific for live vaccinia virus (a smallpox virus surrogate), hen egg lysozyme, cholera toxin, ricin, and staphylococcal enterotoxin B were selected.
