Immunogen
The original antibody was generated by immunizing a llama with CD8, followed by antibody library construction. The antibody was isolated by biopanning against CD8.
Application Notes
The binding affinity of the original format of the antibody to CD8 was measured by surface plasmon resonance (Kd= 0.9 nM). FACS analysis of splenocytes and lymp nodes gated on CD45 + CD19- CD3+ cells, confirming that the original format of the antibody stained CD8+ cells. The antibody competed with a commercial anti-CD8α antibody, showing it bound to the CD8α chain. 89Zr- PEGylated antibody was used for PET imaging on mice, to track the presence of intratumoral CD8+ T cells (Rashidian et al., 2017; PMID: 28666979).
