Immunogen
The original antibody was generated by immunizing alpacas with avian influenza virus nucleoprotein, creating the appropriate phage display library, and panning against the original target.
Application Notes
The binding specificity of the original camelid (alpaca) single-domain antibody (sdAb; VHH) was tested in an ELISA. This antibody was used to generate an anti-avian influenza nucleoprotein and anti-human IgA bi-specific construct together with Ab04649 [A8]. The bi-specific construct detected a range of influenza virus strains in ELISA, including low-virulence and vaccine strains. Namely, H1N1, H3N2, H5N3, H5N1 (LP), H7N3(LP), H8N4, H10N8, H11N6, H12N5, H2N2, H4N6, H5N9, H6N2, H7N7, H9N2, H10N7,H12N6, and H13N6. The bi-specific construct also exhibited a significantly prolonged stability in vivo in Balb/c mice compared to the sdAb version of the antibody (CN111116752B).
