Specificity
This antibody specifically binds an epitope in domain III of the human ST2L and cross reacts with cynomolgus monkey ST2L. ST2L a transmembrane receptor that belongs to the IL-1 receptor family which is expressed on the cell surface of a wide variety of immune cells including T cell, NK/NKT cells, basophils, eosinophils, mast cells and the newly-described non-B/non-T innate lymphoid type 2 cells, nuocytes, and natural helper cells. ST2L expression is also inducible on dendritic cells (DCs), macrophages, and neutrophils. ST2L is able to downregulate the responsiveness of Toll-like Receptors TLR2, TLR4, and TLR9, but also induce type 2 cytokine release via activation by its ligand IL-33 and association with accessory protein IL-1RAcP.

Immunogen
The original antibody was isolated from a human Fab phage display library by panning against hST2L ECD (residues 18–328)-Fc fusion protein. Later on the antibody was converted to a human IgG2 format.
Application Notes
This antibody is not capable of blocking the interaction between IL-33 and ST2L. This antibody was capable of recognizing recombinant human ST2L extracellular domain (ST2L-ECD) using an ELISA assay (PMID: 27294560). The original human IgG2 chimeric version of the antibody binds human and cynomolgus monkey ST2L with a binding affinity of 38 pM and 76 pM respectively (US9090694). This antibody was capable of binding ST2L expressed by LAD2 mast cells in a flow cytometric assay. This antibody was reported to block the IL-33 induced activation of NF-kB in a dose dependent manner. This antibody also blocked IL-1RAcP from binding to the ST2L/IL-33 complex. It can also inhibit cytokine secretion by human basophilic cell line KU812 (PMID: 27294560).
