Immunogen
The original antibody was generated by immunizing dromedary with the enzyme and isolating specific binders from the VHH cloned from peripheral blood lymphocytes.
Application Notes
This antibody can bind porcine pancreatic alpha-amylase in an ELISA and inibits the enzyme with an IC50 = 10nM (PMID: 9649422). This antibody interacts with the amino acid residues in the alpha amylase catalytic site and has a strong inhibitary action on the catalytic activity of the enzyme. The inhibitory interaction of AMD9 involved mainly its complementarity-determining region (CDR) 2 loop, whereas the CDR3 loop was small and certainly did not protrude (PMID: 11960990).
