Immunogen | This conformation specific synthetic antibody was generated by utilizing a phage display library wherein the the Fab fragments is fused to the minor coat protein pIII of the M13 bacteriophage. To generate sABs that preferentially bind to the closed form of MBP (closed-specific sABs), the selection was carried out in the presence of 10 μM maltose. After fours rounds of selection the isolated colonies were subjected to competitive phage ELISA to provide an estimate of affinity towards MBP and specificity for closed form. |
