Key features and details | |
Cat. No. | MABL-3434 |
Name | Anti-MBP mAbs [sAb-11M](MABL-3435) |
Clone No. | sAb-11M |
From | Recombinant Antibody |
Isotype | Engineer antibody |
Application | ELISA |
Species Reactivity | Escherichia coli |
Basic Information | |
Specificity | This antibody is a conformation specific antibody and binds the maltose binding protein in its closed conformation. This antibody is reported to binf the hinge region of the MBP which lies in the opposite direction of the maltose-binding pocket. |
Alternative Name | malE; MMBP; Maltose-binding periplasmic protein; Maltodextrin-binding protein |
UniProt | P0AEY0; P0AEX9 |
Immunogen | This conformationally selective synthetic antibody was generated by customized phage display selections that stabilize MBP in different conformational states. |
Application Notes | This antibody binds MBP with affinity of 20nM in presence of 1 µM maltose. Upon binding to MBP, sAB-11M shifts the MBP conformation to the closed form, thereby covering/altering the epitope and preventing the binding of antibodies specific for open confirmation of the MBP (e.g. sAB-7O). It is reported that in the presence of a 5-fold excess of sAB-11M increases the affinity for maltose by 120-fold. Competitive ELISA was used to determine that the open (sAB-7O) and closed conformation (sAB-11M) specific antibodies compete for binding site of MBP (PMID: 29321208). |
