Specificity | This antibody reacts specifically with physiologically active 2-cis-(S)-form of the endogenous plant growth regulator, abscisic acid. This antibody does not cross react with ABA-catabolites, phasic and dihydrophaseic acid, (R)-ABA, 2-trans-ABA, the ABA-conjugate, ABA-P-D-glucopyranosyl ester, putative ABA precursor and xanthoxin. Abscisic acid is an important sesquiterpenoid plant hormone which acts as a regulator of plant responses to environmental stresses including drought, soil salinity, cold tolerance, freezing tolerance, heat stress and heavy metal ion tolerance. It also plays a role in many plant developmental processes. |

Immunogen | The original antibody was generated by immunizing BALB/c mice with an antigen was which was synthesized by reacting 2-cis-(R, S)-ABA4’-tyrosylhydrazone with p-aminohippuric acid-substituted bovine serum albumin emulsified in an equal volume of complete Freund’s adjuvant. |
Application Notes | The binding affinity and specificity of the original IgG1 antibody was determined using radioimmunoassay (Mertens et al., 1983). This antibody was also used in the generation of a highly sensitive enzyme amplified immunoassay to detect plant growth regulator abscisic acid in homogeneous tissue samples dissected from Vicia faba L. leaves (PMID: 16593922). This antibody was also used in a study to detect whether it can identify any abscisic acid analogs and it was found that this antibody did not cross-react with analogs with the ring double bond reduced and the C-2' methyl cis to the sidechain (PMID: 16667979). An scFv version of this antibody was generated using phage display technology. Specificity of this scFv antibody to abscisic acid was comparable to the binding properties of the complete IgG1 antibody which was verified by competition ELISA (Artsaenko et al., 1994; pmid:8528285). The binding kinetics of the scFv version and the whole IgG1 parental version of this was studied in PMID: 27023768. |
