Immunogen | The original antibody was raised by immunizing a mouse with purified A2AAR-antagonist ZM241385 complex 3. |
Application Notes | The original antibody was used for an ELISA, where it showed it was able to bind its antigen. The antibody was used for a ligand binding assay. Here it showed that the antibody was able to stop the binding of agonist NECA but not that of antagonist ZM241385. This suggests that this antibody induces a inactive confirmation of A2AAR. The structure of the original antibody was determined using x-ray crystallography (Hino et al., 2012; PMID:22286059). The heavy chain of this antibody was cloned into a plasmid and HEK293T were transfected. In vitro is showed it was able to block the A2AR signalling pathway (Zhang et al., 2019; PMID:32308059). The protein-protein interaction of this antibody with its antigen was assessed using mCBB CN-PAGE (Suzuki et al., 2018; PMID:29432751). |
