Anti-PAI-1 mAbs

简要描述:

This antibody binds the reactive center loop (RCL) of human PAI-1 and at the same exosite used by both tissue and urokinase plasminogen activators (tPA and uPA). This antibody binds the active form of human PAI-1. Plasminogen activator inhibitor-1 (PAI-1), is a serine protease inhibitor, a unique member of the serpin superfamily which plays an important role in fibrinolysis and is an established risk factor for cardiovascular diseases. It a primary inhibitor of tissue-type plasminogen activator (PLAT) and urokinase-type plasminogen activator (PLAU). As PLAT inhibitor, it is required for fibrinolysis down-regulation and is responsible for the controlled degradation of blood clots.

Basic Information


Specificity

Immunogen

The original antibody was isolated from a   human scFv library by panning on recombinant human and rat PAI-1 followed by   screening in a PAI-1/tPA competition binding assay.

Application Notes

This antibody specifically inhibits serine   protease interactions with PAI-1 while conserving vitronectin binding. The   binding of this antibody to native and latent form of human PAI-1 was   determined using ELISA. This antibody was able to neutralize recombinant   human, rat and mouse PAI-1 activity in species-relevant tPA-coupled   chromogenic assays. The Fab version of this antibody binds human and rat   PAI-1 with a binding affinity (Kd) of 5.34 pM and 84 pM respectively.   Competition binding experiments also demonstrates that MEDI-579 does not   interact with other circulating serpins (PMID: 30733557). This antibody is   effective in slowing the progression of diabetic nephropathy in db/db mice   and that the effect is additive to ACEI (PMID: 27511457).
