Application Notes | The original human IgM antibody’s reactivity was confirmed through various immunoassays, including immune adherence (IA) and immune adherence absorption (IAA) assays. It exhibited strong binding to gangliosides and effectively detected a range of human tumor tissues, such as melanoma, colon adenocarcinomas, and lung adenocarcinomas. The antibody demonstrated significant cytotoxic activity against antigen-positive tumor cells in the presence of complement. It showed notable reactivity with melanoma (62% positive) and lung cancer (100% positive) while showing less reactivity with other cancers. Neuraminidase treatment effectively inhibited its binding, affirming its specificity for gangliosides. The antibody has been used both therapeutically and diagnostically, showing efficacy in treating cutaneous malignant melanoma through intralesional injection, resulting in tumor necrosis and regression. |
