Key features and details | |
Cat. No. | MABL-1607 |
Name | Anti-Flavocytochrome B2 mAbs |
Clone No. | AFD- B2B4 |
From | Recombinant Antibody |
Isotype | Engineer antibody |
Application | ELISA |
Species Reactivity | Saccharomyces cerevisiae |
Basic Information | |
Specificity | The antibody recognized a conformational epitope on the Heme-binding domain of Flb2 (residues 1-99). Specifically, the epitope consists of Asn30, residues 63 to 65 (GluPro-Leu) and 67 to 70 (Ala- Pro-Asn-Val), Asp72, Tyr74, and the two heme propionate groups. |
Alternative Name | Cytochrome b2 mitochondrial; L-lactate dehydrogenase [Cytochrome]; L- lactate ferricytochrome C oxidoreductase; L-LCR |
UniProt | P00175 |
Immunogen | The original antibody was generated by immunizing BALB/c mice with fully purified Flb. |
Application Notes | ELISA indicated that the antibody, either the IgG or the Fab fragment, was able to recognize the heme-binding domain of flavocytochrome b2. (PMID: 8674650) For the Fab fragment, Kd values of 80 nM and 30 nM were determined in ELISA competition experiments and fluorescence quenching titrations, respectively. (PMID: 9521665) |
Antibody First Published | Lé et al. Functional uncoupling between flavin and heme domains in flavocytochrome b2 by a monoclonal antibody. Biochem Soc Trans. 1996 Feb;24(1):16S. PMID:8674650 |
Note on publication | The paper describe the generation of the antibody B2B4, which inhibits the enzymatic activity of flavocytochrome b2. Further studies were carried out using either the purified divalent IgG1 or the monovalent Fab fragment of the antibody. |
