Specificity | This antibody binds human desmoglein-3 between amino acids 1-161 and does not cross react with desmoglein-1. It is a component of intercellular desmosome junctions. It is involved in the interaction of plaque proteins and intermediate filaments mediating cell-cell adhesion. Pemphigus vulgaris (PV) is a potentially lethal skin disease in which epidermal blisters occur as the result of the loss of cell-cell adhesion caused by the action of autoantibodies against desmoglein 3. It is characterized by suprabasal blistering of skin and mucous membranes. |

Application Notes | This antibody can be used for the determination of desmoglein-3 using ELISA. The binding affinity of IgG1 version of this antibody for human desmoglein-3 was determined using surface plasmon resonance and was found to be Kd = 1.77 nM (PMID: 24942562). This antibody was also used in the direct and indirect immunofluorescence staining of frozen human skin sections (PMID: 24942562; 27304671) The IgG1 and IgG4 variants of this antibody induce loss of cell surface Dsg3 in primary human epidermal keratinocytes and suprabasal blisters in human skin (PMID: 27304671). This antibody is pathogenic in nature and caused suprabasal blisters in human skin at comparable doses of 25 μg and 50 μg (PMID: 27304671). |
