The original antibody was selected from a camel-derived nanobody phage display library. A camel was immunized with UV-inactivated purified elementary particles and outer membrane protein complexes of Chlamydia trachomatis (Bu-434 strain) without LPS. The nanobody was selected from the library by screening against the two immobilized C. trachomatis antigens.
Application Notes
The original antibody (camel VHH) was screened for affinity to its target using ELISA. Specific binding to free antigens and the detection of C. trachomatis in eukaryotic (McCoy) cells were visualized via immunofluorescence microscopy. Preincubation of C. trachomatis elementary bodies with this antibody reduced infection in McCoy cell cultures. At higher concentrations (5-10 μg/ml), the nanoantibody significantly decreased both the number and size of intracellular inclusions (US9464132B2).
