The original antibody was generated from phage display libraries constructed using healthy human blood of 40 doners and cord blood of 10 doners and biopanning them against recombinant 5T4-Fc fusion protein conjugated to magnetic beads.
Application Notes
5T4 is an oncofetal antigen expressed in a number of carcinomas including colorectal, ovarian, renal, non-small cell lung and gastric carcinomas, while its expression in normal adult tissues is limited. This antibody is recomended for diagnosis of cancer in a patient. Fab version of this antibody bound to 5T4 with an EC50 of 2 nmol/L, as measured by ELISA. This antibody was also capable of binding native 5T4 associated with cell surfaces. The binding of m1001 to surface expressed 5T4 on A498 cells was confirmed using flow cytometry (WO2016022939).
