Specificity
This antibody binds human Interleukin-1 beta, which is a potent pro-inflammatory cytokine. Initially discovered as the major endogenous pyrogen, induces prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production. It promotes Th17 differentiation of T-cells and synergizes with IL12/interleukin-12 to induce IFNG synthesis from T-helper 1 (Th1) cells. It also plays a role in angiogenesis by inducing VEGF production synergistically with TNF and IL6.

Immunogen
The original mouse antibody was generated by immunizing mice with human IL-1B protein. The humanized version was generated by grafting CDRs of the original mouse antibody onto human framework regions.
Application Notes
This antibody is the humanized version of original mouse antibody IC8. This antibody binds human IL-1B with a binding affinity of Kd= 0.28 nM. The Fab version of this antibody was reported to neutralize IL-1B activity both in an in vitro and an in vivo assay (US7608694).
