Specificity
This antibody binds human CIP2A (cancerous inhibitor of protein phosphatase 2A). CIP2A is an oncoprotein that inhibits protein phosphatase 2A (PP2A), stabilizes MYC, and is involved in the progression of several human malignancies. It promotes anchorage-independent cell growth and tumor formation.Antibodies against CIP2A are present in sera from many patients with gastric or prostate cancer and acts as prognostic marker in many cancers.

Immunogen
The original monoclonal antibody was generated by collecting spleen cells from an immunized mouse, followed by an affinty purification on a goat anti-CSAp IgG column and susequent fusion with the nonsecreting mouse myeloma cell line SP2/0-Agl4.
Application Notes
This antibody binds a single site epitope of the CSAp and has an afiinity constant Ka =9.61 x 108/mol. This antibody is not suitable for sandwich ELISA as it binds to a single site epitope of the CSAp, it is however suitable for direct and competition ELISA. This antibody does not react with reduced and alkylated CSAp. This antibody cannot fully block the reactivity of goat anti-CSAp. Immunohistochemical staining of neoplastic tissues showed the Mu-9-reactive determinant to be present in 60% of colon carcinomas. This antibody showed no reactivity outside the intestinal tract and no reactivity with blood cells (PMID: 2169346). The comparison of therapeutic efficacy and host toxicity of two different 131I-labelled antibodies effects of antibodies, Mu-9 and NP-4 IgG (anti-CEA antibody) revealed that even though the anti tumor effects of both these antibodies are equal, there is a significant difference in their therapeutic effectiveness (PMID: 2759735). Radioimmunotherapy of GW-39 human colonic tumor xenografts grown in the hamster cheek pouch with131I-labeled NP-4 anti-(carcinoembryonic antigen) (CEA) and131I-labeled Mu- 9 anti-(color-specific antigen-p) (CSAp) murine monoclonal antibodies, administered in combination, was more effective than using either antibody alone for tumor masses less than 0.5 cm3 in size (PMID: 1652355).
